Comparison of the performance of anion exchange membrane materials for adenovirus purification using laterally-fed membrane chromatography

Viruses are an emerging class of biotherapeutics used to treat a variety diseases. Ion-exchange membrane chromatography is commonly for virus purification, with the Mustang Q (Pall) and Sartobind (Sartorius) being two most widely strong anion-exchange membranes. These membranes made from different materials have pore sizes, while previous studies attempted compare their performance in various purification applications, those been hindered by additional fact that these sold module formats. Thus, order establish direct comparison capabilities anion exchange materials, we first fabricated equivalent laterally-fed (LFMC) devices containing either 1 mL or then compared based on residence times, elution patterns pre-purified biomolecules, recovery purity adenovirus cell lysate. Under same linear gradient conditions, purified eluted approximately one volume earlier membrane; similar result was obtained during infected suspension 293 cells. While total both (~60%), provided much greater removal DNA impurities fractions.
